Drontal (praziquantel/pyrantel pamoate) tablets will remove Tapeworms (Dipylidium caninum, Taenia taeniaeformis), Hookworms (Ancylostoma tubaeforme), and Large Roundworms (Toxocara cati) in cats and kittens. The presence of tapeworms is indicated by the observance of tapeworm segments passed in the cat's feces. Tapeworm segments are white, pinkish-white or yellow-white in color and are similar in size and shape to flattened grains of rice. The segments are most frequently observed lying on a freshly passed stool. Segments may also be found on the hair around the anus of the animal or on the animal's bedding. Cats become infected with tapeworms after eating fleas or small mammals (rabbits, mice) which may be infected with tapeworm larvae. Hookworms are small whitish or reddish-brown worms less than one inch in length that live in the intestinal tract and feed on blood. Cats can become infected with hookworms by swallowing infected larvae while grooming or when larvae from the environment burrow through the skin. Cats infected with hookworms may have poor physical condition, dull haircoat, and reduced body weight and diarrhea, sometimes with the presence of dark blood. Large roundworms are white or yellow-white strands 2-7 inches in length (similar in size and color to smooth strings of spaghetti) that may be observed in the cat's vomit or feces. Cats become infected with large roundworms by swallowing infective eggs, particularly while grooming, or by ingestion of mice that may be infected with larval stages. Large roundworms and hookworms pass eggs in the feces of the cat that can only be observed with the aid of a microscope. The presence of these parasites should be confirmed through identification of parasite eggs in the feces. Mechanism of Action Credelio Quattro contains four active pharmaceutical ingredients, lotilaner, moxidectin, praziquantel, and pyrantel (as pamoate salt). Lotilaner is an ectoparasiticide belonging to the isoxazoline group. Lotilaner inhibits insect and acarine gamma-aminobutyric acid (GABA)-gated chloride channels. This inhibition blocks the transfer of chloride ions across cell membranes, which results in uncontrolled neuromuscular activity leading to death of insects and acarines. The selective toxicity of lotilaner between insects and acarines and mammals may be inferred by the differential sensitivity of the insects and acarines’ GABA receptors versus mammalian GABA receptors. Moxidectin is an endectocide in the macrocyclic lactone class. Moxidectin acts by interfering with the chloride channel-mediated neurotransmission in the parasite. This results in paralysis and death of the parasite. Praziquantel’s mode of action is not precisely known, but treated tapeworms undergo muscular paralysis accompanied by a rapid influx of calcium ions and the disruption of the tegument. Pyrantel is a nematocide belonging to the tetrahydropyrimidine class. Pyrantel acts as a depolarizing, neuromuscular-blocking agent in susceptible parasites, causing paralysis and death or expulsion of the parasite. Pharmacokinetics Due to reduced drug bioavailability of lotilaner in the fasted state, Credelio Quattro must be administered with a meal or within 30 minutes after feeding. Following a single oral administration of Credelio Quattro at the minimum labeled dose in the fed state or a single intravenous administration of 5 mg lotilaner, 0.005 mg moxidectin, 1.25 mg praziquantel, and 1.25 mg pyrantel per kg body weight to Beagle dogs (1.5 to 2.5 years old), the mean oral bioavailability for lotilaner, praziquantel, and pyrantel was 93%, 41%, and 31%, respectively. Bioavailability for moxidectin is not reported due to insufficient data to adequately describe the elimination phase following intravenous administration. For Credelio Quattro, the area under the curve from time of dosing to the time of the last measurable concentration (AUClast) was 5970, 1.75, 1.54, and 0.857 mg*h/L for lotilaner, moxidectin, praziquantel, and pyrantel, respectively. Peak concentrations (Cmax) of 9070, 15.3, 390, and 122 ng/mL were reached (Tmax) 10, 4.5, 0.75, and 2.5 h after dosing for lotilaner, moxidectin, praziquantel, and pyrantel, respectively. Mean plasma elimination half‐lives were 806, 634, 3.64, and 4.87 h for lotilaner, moxidectin, praziquantel, and pyrantel, respectively. Following nine oral administrations of Credelio Quattro at 1X, 3X, and 5X the maximum labeled dose of 40 mg/kg lotilaner, 0.04 mg/kg moxidectin, 10 mg/kg praziquantel, and 10 mg/kg pyrantel, every 28 days in 8-week-old Beagle dogs, moxidectin and lotilaner area under the curve from time of dosing to the time of the last measurable concentration (AUClast) increased approximately in a less than proportional manner, whereas praziquantel AUClast increased approximately in a more than proportional manner from 1X to 5X after most study doses. Pyrantel AUClast increased approximately in a proportional manner from 1X to 5X observed after first, sixth, and last doses. Within the 1X group, accumulation was observed between Days 0 and 224 with geometric mean accumulation ratios for AUClast of 6.2 and 7.9 for lotilaner and moxidectin, respectively. Concentrations of praziquantel and pyrantel prior to each dose were below the limit of quantification.

Parasite resistance may develop to any dewormer, and has been reported for most classes of dewormers. Treatment with a dewormer used in conjunction with parasite management practices appropriate to the geographic area and the animal(s) to be treated may slow the development of parasite resistance. Fecal examinations or other diagnostic tests and parasite management history should be used to determine if the product is appropriate for the herd/flock, prior to the use of any dewormer. Following the use of any dewormer, effectiveness of treatment should be monitored (for example, with the use of a fecal egg count reduction test or another appropriate method). A decrease in a drug's effectiveness over time as calculated by fecal egg count reduction tests may indicate the development of resistance to the dewormer administered. In a field safety and effectiveness study, Credelio Quattro was administered to dogs for the prevention of heartworm disease. The study included a total of 372 dogs treated once monthly for up to 11 treatments (191 treated with Credelio Quattro and 181 treated with an active control). Over the 330-day study period, all observations of potential adverse reactions were recorded. Adverse reactions seen during the field study are summarized in the table below. The dog recovered within 24 hours after the first episode and 1 hour after the second episode and completed the study. One dog receiving Credelio Quattro was observed by the investigator to have anisocoria during scheduled physical examinations one month after the ninth dose and one month after the eleventh dose. In a second U.S. field safety and effectiveness study, Credelio Quattro was administered to 103 dogs with hookworms. Adverse reactions in dogs treated with Credelio Quattro included gastrointestinal signs (diarrhea, emesis) and lethargy. In a U.S. field study, 165 dogs received a combination of lotilaner, moxidectin, and praziquantel, three of the active ingredients at the same doses as in Credelio Quattro, monthly for up to 11 months. Two dogs with no history of seizures experienced seizures during the study. One of the dogs developed cluster seizures and was removed from the study. Ataxia was also observed in one other dog three days after the first dose. In a U.S. field study, one dog administered lotilaner alone, a component of Credelio Quattro, was observed with intermittent head tremors within 1.5 hours of administration of vaccines, an ear cleaning performed by the owner, and its first dose of lotilaner. The head tremors resolved within 24 hours without treatment. The owner elected to withdraw the dog from the study. In an Australian field study, one dog with a history of seizures experienced seizure activity (tremors and glazed eyes) six days after receiving lotilaner alone. The dog recovered without treatment and completed the study. In a U.S. field study, two dogs with a history of seizures received lotilaner alone and experienced no seizures throughout the study.

Credelio Quattro™ CredelioQuattro™-CA1(lotilaner, moxidectin, praziquantel, and pyrantel chewable tablets)

Field study: In a controlled, double-masked field safety study, owners administered PROFENDER Topical Solution to 606 cats. Adverse reactions reported by the cat owners included licking/excessive grooming in 18 cats (3.0%), scratching treatment site in 15 cats (2.5%), salivation in 10 cats (1.7%), lethargy in 10 cats (1.7%), alopecia in 8 cats (1.3%), agitation/nervousness in 7 cats (1.2%), vomiting in 6 cats (1.0%), diarrhea in 3 cats (0.5%), eye irritation in 3 cats (0.5%), respiratory irritation in 1 cat (0.2%) and shaking/tremors in 1 cat (0.2%). All adverse reactions were self-limiting. Laboratory effectiveness studies: One cat died 10 days after receiving PROFENDER Topical Solution. The necropsy showed chronic active cholangiohepatitis. While the use of the drug did not appear to be the direct cause of death, treatment with the drug cannot be ruled out as a contributing factor (See PRECAUTIONS ). One cat treated with a vehicle placebo (formulation minus the active ingredients) showed salivation, gagging, lethargy and a swollen tongue. Foreign Market Experience: The following adverse events were reported voluntarily during post-approval use of the product in foreign markets: application site reaction (hair loss, dermatitis, pyoderma, edema, and erythema), salivation, pruritus, lethargy, vomiting, diarrhea, dehydration, ataxia, loss of appetite, facial swelling, rear leg paresis, seizures, hyperesthesia, twitching, and death. Topical Solution profender™ (emodepside/praziquantel)

In a field safety and effectiveness study, which included a total of 201 households and 380 treated cats (244 cats treated with NexGard® COMBO, 136 cats treated with an active control), the safety of NexGard® COMBO was evaluated over a 90-day period through in-clinic physical examinations or through reporting of abnormalities by the owner. The most frequently reported reactions in the NexGard® COMBO and active control groups are presented in the following table. NexGard® COMBO (esafoxalaner, eprinomectin, and praziquantel topical solution)

In a field study with IVERHART MAX Chew, self-limiting adverse reactions, including vomiting, diarrhea, lethargy, difficulty swallowing, excessive salivation, increased water consumption, and coughing were reported. Self-limiting adverse reactions, including lethargy, limpness, salivation, shaking, diarrhea, decreased appetite, licking lips, and belching were reported between 20 minutes and 72 hours following treatment in a field study with ivermectin/pyrantel pamoate/praziquantel tablets. In field studies with ivermectin/pyrantel/praziquantel pamoate tablets, vomiting or diarrhea within 24 hours of dosing was rarely observed (1.1% of administered doses). IVERHART MAX® Chew (ivermectin/pyrantel pamoate/praziquantel)

Mild side effects were observed in 18 of 189 dogs (9.5%) and 8 of 85 cats (9.4%) administered Praziquantel Injection in field trials. For dogs the majority of these were described as brief pain responses following injections to larger dogs (weighing over 50 lbs.) Two dogs exhibited a brief period of mild vomiting and/or drowsy or staggering gait. The eight cats exhibited either diarrhea, weakness, vomition, salivation, sleepiness, burning on injection and/or a temporary lack of appetite. Local irritation or swelling at the site of subcutaneous injections have been reported for cats. CAUTION: Federal law restricts this drug to use by or on the order of a licensed veterinarian.

Place the tip of the applicator on the skin and apply the entire contents directly onto the skin in one spot. The product should be applied to dry skin on an area where the cat cannot lick it off. If the weight of the cat requires a second application, apply the contents in the same manner as described above in the same location. 6.Wash hands after use with soap and water. Heartworm Prevention: For the prevention of heartworm disease, NexGard® COMBO should be administered once a month year-round. Flea Treatment and Prevention: For the treatment and prevention of flea infestations, the use of NexGard® COMBO may begin at any time of year. However, an environmental infestation may persist for a short time after beginning treatment with NexGard® COMBO because of the development of adult fleas from eggs that were laid prior to the initiation of treatment. Credelio Quattro/Credelio Quattro-CA1 is given orally at the minimum dosage of 9 mg/lb (20 mg/kg) lotilaner, 0.009 mg/lb (0.02 mg/kg) moxidectin, 2.28 mg/lb (5 mg/kg) praziquantel, and 2.28 mg/lb (5 mg/kg) pyrantel (as pamoate salt). When replacing another heartworm preventative product in a heartworm disease prevention program, the first dose of IVERHART MAX Chew must be given within a month (30 days) after the last dose of the former medication. A heartworm test should be performed prior to and 6 months after switching heartworm preventative products. If the interval between doses exceeds a month (30 days), the effectiveness of ivermectin can be reduced. Therefore, for optimal performance, the chew must be given once a month on or about the same day of the month. If treatment is delayed, whether by a few days or many, immediate treatment with IVERHART MAX Chew and resumption of the recommended dosing regimen will minimize the opportunity for the development of adult heartworms.

IVERHART MAX® Chew (ivermectin/pyrantel pamoate/praziquantel)

This syringe contains sufficient paste to treat one 1320-lb horse at the recommended dose rate of 91 mcg ivermectin per lb (200 mcg/kg) and 0.68 mg praziquantel per lb (1.5 mg/kg) of body weight. Each weight marking on the syringe plunger delivers enough paste to treat 220 lb (100 kg) of body weight. 1. While holding plunger, turn the knurled ring on the plunger 1/4 turn to the left and slide it so the side nearest the barrel is at the prescribed weight marking. 2. Lock the ring in place by making a 1/4 turn to the right. 3. Make sure that the horse’s mouth contains no feed. 4. Remove the cover from the tip of the syringe. 5. Insert the syringe tip into the horse’s mouth at the space between the teeth. 6. Depress the plunger as far as it will go, depositing paste on the back of the tongue. 7. Immediately raise the horse’s head for a few seconds after dosing. Do not underdose. Ensure each animal receives a complete dose based on a current body weight. Underdosing may result in ineffective treatment, and encourage the development of parasite resistance. Parasite Control Program: All horses should be included in a regular parasite control program with particular attention being paid to mares, foals, and yearlings. Foals should be treated initially at 4 weeks of age, and routine treatment repeated as appropriate. Consult your veterinarian for a control program to meet your specific needs. Replace 3/4 of the aquarium water prior to beginning treatment with dactymor. Add 5 ml (= 100 drops) of dactymor for every 100 l of aquarium water and stir the aquarium water to disperse the product. Switch off UV-lamps. After 6 hours treatment a replacement of minimum 3/4 of the aquarium water is required. On the following day change half of the aquarium water. A single treatment is generally adequate for skin worms and tapeworms. However, trematode eggs are not affected. Therefore, when treating trematodes, the treatment should be repeated twice, waiting approx. one week between each treatment (for coldwater ornamental fish, wait approx. one-and-one-half weeks between treatments). The aquarium should be continuously monitored during the treatment with dactymor. In aquariums containing a large biomass (microorganisms in filters, ground, etc.) the use of dactymor may result in an increase in bacterial growth. In turn this can lead to an oxygen depletion. Immediately change the aquarium water if signs of bacterial turbidity or indications of oxygen depletion are observed.

Virbantel® Flavored Chewables

1) While holding plunger, turn the knurled ring on the plunger ¼ turn to the left and slide it so the side nearest the barrel is at the prescribed weight marking, aligning the arrow on the plunger with the notch on the ring, as shown in the pictogram. 2) Lock the ring in place by making ¼ turn to the right. Ensure it is locked (it should no longer slide). 3) Make sure that the horse's mouth contains no feed. 4) Remove the cover from the tip of the syringe. 5) Insert the syringe tip into the horse's mouth at the space between the teeth. 6) Depress the plunger as far as it will go, depositing paste on the back of the tongue. 7) Immediately raise the horse's head for a few seconds after dosing.

Droncit (praziquantel) Injectable Cestocide may be administered by either the subcutaneous or intramuscular route to dogs and cats. The intramuscular route may be preferred in dogs due to a brief period of pain that occasionally follows subcutaneous administration. Anaphylactoid reactions were not observed in clinical trials. However, as with any drug an anaphylactoid reaction can occur with this product and should be treated symptomatically if it occurs.
